(1) Reason: The amount of adhesive or lubricant is small or inappropriate, resulting in uneven distribution of particles, and stratification of coarse particles and fine particles. Even increasing the pressure during tablet pressing cannot overcome this problem.
Solution: You can choose appropriate adhesives or increase the amount, improve the granulation process, mix the particles evenly, etc.
(2) Reason: The drug is not ground fine enough, and the content of fibrous, elastic drugs or oil components is high, resulting in uneven mixing.
Solution: You can grind the drugs into smaller pieces, choose adhesives with strong viscosity, increase the pressure of the tablet press, add oil-based drug absorbents, and mix them thoroughly.
(3) Reason: The water content is not moderate. Too little water or the dried particles are elastic. Because the drugs containing crystalline water lose more crystalline water during the drying of the particles, they become brittle and easy to crack. But too much water will reduce the hardness.
Solution: The water content should be controlled according to different varieties during the granulation process. If the granules are too dry, spray an appropriate amount of dilute ethanol (50%-60%), mix well and press into tablets.
(4) Cause: Physical properties of the drug itself. Determined by brittleness, plasticity, elasticity and hardness. For example, elastic materials become smaller when compressed and expand due to elasticity after decompression, so the tablets are loose and cracked.
Solution: Different drugs require different pressures when tableting and other auxiliary materials are added for control.
(5) Cause: Mechanical factors. For example, the punch length is uneven, or the pressure adjustment is inappropriate, the tablet press speed is too fast or the particles in the hopper are sometimes more and sometimes less.
Solution: The tablet press pressure can be adjusted, the punch, tablet press speed and feeding speed can be replaced.
